Preparation: Add 0.3 mol aniline, 40 mL glacial acetic acid and 0.4 g zinc powder to a round-bottomed flask, install a fractionating column, install a thermometer on the top of the column, and collect the distilled water and acetic acid in the beaker. Heat slowly for 15 minutes, then distill at around 105°C. When the temperature continues to drop, stop heating and reach the end point. Pour the reactant into ice water under stirring conditions while it is hot to precipitate a solid, cool and filter with suction to obtain crude acetanilide, which can be recrystallized with water to obtain pure white 2-methylacetanilide. Add 0.05 mol of the product 2-methylacetanilide and 0.15 mol of DMF obtained in the previous step into a four-necked flask, install a thermometer, mechanical stirrer, and condenser tube, stir mechanically in an ice bath, and keep at a temperature lower than 10°C. Slowly add 0.35 mol POCl3 dropwise. After the dropwise addition is completed, raise the temperature to 75°C. Stir and react for 12 hours. The color gradually changes from light yellow oily liquid to orange-red liquid. After the reaction is complete, stop heating and slowly stir the reactant. Pour into 400 mL ice water, let it stand, filter the precipitated solid, wash it three times with water, dry it in the air, and recrystallize it with ethyl acetate to obtain light yellow needle-like crystals 2-chloro-8-methylquinoline-3. -formaldehyde. 2-Chloro-8-methylquinoline-3-carbaldehyde prepared by acetanilide cyclization synthesis method is characterized by mass spectrometry, elemental analysis, infrared, nuclear magnetic resonance spectroscopy, etc. as follows: light yellow needle-shaped crystals, yield 90% , mp.150~152℃.
